1) To check amplicon oresent
2) PCR clean up
3) restriction digestion,
4) ligation, SLiCE, or gibson assembly steps
5) much better to do chemical transformation instead of electroporation when cloning. Otherwise, forgot to dialyse before electroporation

@MicrobiomDigest I would think corresponding authors conceived the study and wrote the paper is not that unusual, although more normal if it was your own lab doing the experiments for you to write the paper
