MIBIREM partner @Lesaffre_Group — Notes from the lab 🧪
After enrichment cultures of cyanide-degraders were established, we now use Adaptive Laboratory Evolution (ALE) in a fully automated system to further improve these microbiomes.
Starting culture: a community adapted to hexacyanoferrate(II), a common cyanide-iron complex at former gas work sites (less toxic than free cyanide).
Goal: increase degradation rates via continuous selection, with regular sampling (glycerol stocks + DNA analysis). A pre-experiment is now testing growth conditions in the ALE device.

"At the end of February, oxygen injection began at the #Stockach site, marking the start of full-scale remediation of the cyanide groundwater contamination.
This is an in-situ biological remediation measure. A groundwater circulation cell will first be set up in the hot spot area. To stimulate the microbial degradation processes, the inflowing groundwater will be enriched with oxygen.
Once a stable oxygen concentration has been achieved in the groundwater within the circulation cell, glucose will be added as a carbon source. From this point on, the microorganisms have sufficient oxygen and readily available carbon at their disposal to degrade the cyanide by using it as a nitrogen source. In the further course of the project, bioaugmentation with a laboratory culture containing bacterial strains selected for the degradation of complex cyanide compounds is also planned.
As part of the EU research project @mibirem the remediation measure is being monitored by means of close sampling. Samples are taken not only for chemical analysis, but also for the isolation of environmental DNA. The MIBIREM research partners hope that this will provide deep insights and a better understanding of the microbial processes during such a remediation measure, which can then also be used to optimize the remediation." 💫

"After several weeks of on-site work in Ploufragan, the pilot plant has officially been launched on Monday, 9 March. 👏
At the Ploufragan site, 10 piezometers were installed across two areas for extraction, injection, and monitoring, in collaboration with the environmental agency Ademe. An irrigation system was also set up to enhance moisture in the unsaturated soil layers.
The bioflushing plant was then commissioned, consisting of tanks, filters, a bioreactor, and pumps. Groundwater is extracted, treated, and reinjected into the pilot area—enriched with nutrients, oxygen, and biomass to stimulate microbial activity.
Initial (T0) soil and water samples have been collected and sent for chemical and microbial analysis including at CNRS. A dedicated site office has also been installed to support the pilot phase.
Step by step, this work brings us closer to demonstrating effective, microbiome-based solutions for soil and groundwater remediation. "🌱
