Below a cool teaser of what the microscopy setup from our latest preprint can do. Here, a Salmonella bacterium (arrow) divides on the surface of human intestinal cells. It then invades, and the epithelial cell reacts by instantly killing itself.

@pmoyniha@GlcNAcMurNAc I agree with you. We have discussed potential reasons for differences in our observations in the discussion section. Since Tn mutant pool is an outgrowth pool, it is inherently difficult to detect growth defective mutants such as this. But Tn mutants are powerful first screen.
An interesting protein with domains reminiscent of a classical PBP, but it also exhibits a strong b-lactamase activity. M. abscessus mutant lacking this protein grows as a single clump and is wiped out from the lungs of mice...Thx @GlcNAcMurNAc for sharing this.
@FreundlichGroup Thx Joel. A particular finding reported in our 2017 NCB paper, that penems react more strongly with LDTs catalyzed this project. I collaborated with you for several studies reported in that paper. That was also an excellent team effort.
