@RajaFlores I don’t know that we have the answer to this most important of questions yet... But I do know that patients who get a PCR to neoadj chemo-IO have a 95% OS at 3 years, which is better than surgery for stage IA based on @AltorkiNasser and colleagues’ lobe vs sublobe trial.
Similar to Usain Bolt, lung cancer continues to surge past the competition for drug discoveries leading to FDA approvals. #lcsm@usainbolt@FDAOncology https://t.co/fPlP8AlgSr
@HenningWillers@toddscarbrough@MARIANOPROVENCI Effect is broader than pCR, watch this space! Further in depth path analyses to come soon will help elucidate who else is benefiting (and perhaps that emerging breathing room between the non-pCR curves).
Further implication from Dr. @MARIANOPROVENCI presentation #ESMO23 . I don’t envy the statistician designing an adjuvant/perioperative study for pts with lung cancer pCR after neoadjuvant alone - 95% of pts are alive at 3 years. Amazing result for our pts! #lcsm
#ESMO23 In PDL1 negative, outcomes not nearly as good in CheckMate 816. pCR 16.7%, EFS HR 0.87, OS HR 0.81 with 3y OS rate 71% vs 60%. Regardless of that, all who achieved pCR had excellent survival. More lobectomy, fewer pneumonectomy. Congratulations @MARIANOPROVENCI
#ESMO23 Demographics show largely squamous NSCLC in pts with smoking history. Neoadjuvant tislelizumab with a huge improvement in MPR (56.2% vs 15.0%) and in pCR (40.7% vs 5.7%). A 40.7% pCR rate is quite high (blinded pathologic review) - reflecting drug or just pt population.
Rapidly shifting standards & tons of new data in lung cancer at #ESMO23. To process these data & hear from the experts themselves, come to Austin, TX April 18-20 for #TexasLung24 at Austin City Limits @acllive. See faculty & agenda below! @TLCconference

Dr. @MARIANOPROVENCI with 3 yr detailed results on #CM816.
Overall survival PD-L1 pos✅
OS PD-L1 neg - 71% vs. 60% at 3yrs, did EMA speak too soon?
Path complete response ~ OS ✅
That non-pCR curve still favors those 3 cycles of chemo-nivo, maybe a small truck at 5 yrs? #ESMO23
Behind the numbers & data there are real people with a story, and I share this to possibly provide some hope and express my profound gratitude. I was able to celebrate today with my mom because of research, medicine, and MDs who care. Just last Summer she was dx 3a NSCLC, and after resection & a PCR from neoadj chemo/IO, my heart is whole and I am grateful. #checkmate816 #CM816 #lcsm #OncTwitter @BorisSepesi@FordePatrick@DoctorJSpicer @JimAllisonPhD @MDAndersonNews
Dr. @IyengarPuneeth outlines oligometastatic lung cancer and the potential benefit of local therapy. Some important studies with encouraging results. Perhaps more benefit with IO (@Jbauml et al). Important larger studies underway but what is the right endpoint? #PLCClive23
CheckMate 816 - 3 year overall survival data with chemo-immunotherapy given before surgery for lung cancer was presented today, here is the patient-oriented manuscript describing the trial and results! #LCSM
It’s for real folks! If you kill the tumor in the primary and lymph nodes, you’re killing it wherever else it might be. PCR after chemo-IO is a survival surrogate! Now we just need better measures of micromet disease to adjudicate neoadj more precisely. #elcc23@IASLC

3-yr Update of CM816 trial: Neoadj chemoIO v chemo shows
- contd EFS benefit
- OS HR 0.67 (p=0.012)
- *chemoIO appears to protect against CNS progression* - this is the gold
Impressive data. Identifying what further IO adds, is the key quesn

Three year overall survival after neoadjuvant chemo-nivolumab for surgically resectable lung cancer results coming up this Thursday, March 30 at #ELCC23 Results at 2 years below! #LCSM
Neostar strikes again, led by Dr. Tina Cascone! Out now in @NatureMedicine neoadjuvant chemo-ipi-nivo in resectable lung cancer. The exciting study results here https://t.co/bh2eWBJ6ME & commentary from @michaelconroy https://t.co/0eV58TMzbD Exciting times! #LCSM
What term do you use for lung cancer pts who had PCR after neo chemo/IO & surgical resection? NED? Cured? What if they asked you if the were cured? With the evolution of treatment options, there’s more going on than just “stable disease.” I imagine there are differing opinions on this one. #onctwitter #lcsm @DoctorJSpicer@FordePatrick@JackWestMD@StephenVLiu@BrendonStilesMD
A lot of people coming up in data science these days seem to think the point of learning data science is so you can get a fancy data science job.
I think the point is it empowers you to analyze data and come to solid conclusions which is useful no matter what your job is.
