A Cell study of 40 patients reveals two key escape routes.
🧪 KRAS mutations
KRAS Y64 variants weaken drug binding and disrupt complex formation. KRAS Y71 mutations instead strengthen RAS–RAF interaction, competing against the drug effect.
🧬 BRAF alterations
Kinase-dead or low-activity BRAF mutants stabilize RAF dimers, sustaining downstream signaling even under treatment.
🔁 Overall, resistance converges on restoring RAS–RAF signaling—either by weakening drug binding or strengthening native protein interactions.
