Jay found lymphoid structures in the skull bone marrow that seem to be serving immune hub role for the brain... brain, after all, may not be using "common lymph nodes" but rather has its own unique immune hubs and Jay showed their role in brain tumors... but their role in likely much more than that. Moreover, our findings identify IL-1R2 as a novel "adaptokine"—a secreted protein induced by cellular or organismal stress to maintain immune-metabolic homeostasis. 7/
💡 Why it matters: This study strengthens the importance of the BM–liver axis in controlling alcohol-associated inflammation—and suggests translational opportunities, including IL-1R2⁺ monocyte–based cell therapy and recombinant IL-1R2 as potential therapeutic strategies. 6/
👥 Importantly, in patients with chronic alcohol consumption, we also observed increased IL-1R2⁺ monocytes in blood and liver, supporting clinical relevance. 5/
🍺 With chronic alcohol exposure, LepR⁺ bone marrow MSCs metabolize alcohol, experience oxidative stress, and excrete glutamate via the xCT transporter—along with increased chemokine ligands that help recruit immune cells. 1/
