In the final step, our minicells bind specifically to tumor tissue and then inject proteins to inhibit the cancer cell growth. To do this, our minicells will form complexes on their outer shell that bind to specific surface structures of the cancer cells.
Phase 2: Cancer cell adhesion & protein secretion.
The injectisome, a complex inherent to Salmonella, will be used for protein secretion. It has a secretion system and a needle-like structure that protrudes from the cell. The injectisome is also inherent in our minicells.
Our minicells will form complexes on their outer envelope that bind specifically to these surface structures. This will be the most difficult step, but we found promising candidates!

Some information to Phase 1:
We want to derive achromosomal minicells from our attenuated #Salmonella Typhimurium strain to use them as drug delivery system.
There are different approaches to yield bacterial minicells. We are using the disruption of the Min system.
By deletion of the minD gene, cells are unable to restrict the divisome to mid-cell, leading to septum formation at cell poles and constriction of nucleoid-free minicells.
