Bacteria can construct DNA from scratch — without using DNA at all.

For decades, the central dogma of biology has been clear: DNA makes RNA, and RNA makes proteins. To build new DNA, cells must copy an existing DNA or RNA template. However, researchers from Stanford University and Columbia University have shattered this fundamental rule.

While studying how Escherichia coli bacteria defend themselves against viral attacks, scientists uncovered a defense system called DRT3. Inside this system, an enzyme known as Drt3b does the seemingly impossible: it constructs a new complementary strand of DNA entirely without a genetic blueprint. Instead of reading nucleic acids, the enzyme uses its own internal protein structure and amino acid geometry as a physical mold to determine which DNA letters to add.

This unprecedented discovery is the biological equivalent of using a baked cake to write a recipe, rather than the other way around. By allowing information written in amino acids to dictate the creation of new DNA, this protein-templated synthesis proves that genetic code can be generated without copying an existing genome. Beyond rewriting textbook biology, this mechanism opens up immense possibilities for synthetic biology.

Engineers may soon harness these blueprint-free enzymes to manufacture customized DNA strands for advanced biomaterials, targeted gene therapies, and dynamic synthetic cells. It suggests that the origin of genetic diversity and the mechanics of life are far more flexible, and far stranger, than we ever imagined.
