10/ Overall, we demonstrated a synergistic effect of JAK2 mutation and chr9p duplication in MPNs. This condition results in:
1. Higher CD34+ stemness and clonogenicity due to OCT4+NANOG upregulation
2. The establishment of a PD-1/PD-L1 dependent immunosuppressive phenotype
Blockage of CTLA-4, an immune checkpoint receptor on T cells, appeared to boost the cytotoxic T-cell–mediated immune response against #myelofibrosis cells in human xenografts.

@rosslevinemd In the recent ESHMPN meeting our poster showed that chr9p duplication in JAK2mut MPN patients results in higher clonogenicity, stemness and immune evasion. Not a p53-related CNV, but still a relevant proof that CNVs definetely play a disease-modifying role 🧩
