Off-the-shelf AI models are powerful, but they often stumble when facing complex, real-world drug development challenges like antibody polyreactivity—one of the most common reasons promising candidates fail early.
Our latest research published in mAbs details how we used locally tuned in silico AI models to efficiently resolve antibody polyreactivity during drug development.
⚫ Reliable Ground Truth Data: We introduced Range Normalized Summation (RNS) scores derived from multi-concentration mAb ELISAs. Demonstrating exceptional reproducibility across different reagents and separate experiments, these scores provide the high-quality ground truth measurements required to build expandable datasets for modeling.
⚫The AI Bridge (Fine-Tuning PLMs): To translate this data into better antibody designs, we train AI specifically by fine-tuning Protein Language Models (PLMs) on localized sequence data. We demonstrate specialized tuning is required for efficiently inferring novel, low-, or non-polyreactive variants in some contexts, in this case driving a 66% success rate with just 18 attempts vs 0% for the top performing base model.
⚫ Transparency & Advancing the Field: Antibody sequences and associated in vitro polyreactivity scores are provided in the supplemental material for use in future benchmarking studies.

Key Highlights:
- Study medication appears well tolerated, with favorable safety data across all blinded single ascending dose (SAD) cohorts
- Estimated half-life of at least 65 days supports potential for ABS-201 targeted dosing interval of two or three injections over six-month period
- First multiple ascending dose (MAD) of ABS-201 in cohort of androgenetic alopecia participants has been initiated
It's a strong start for ABS-201, and we're looking forward to interim proof-of-concept data in the second half of 2026, with full PoC data in early 2027.

We’re excited to introduce Origin-1: a generative AI platform for de novo antibody design against "zero-prior" epitopes.
Origin-1 has been successfully validated against four targets: COL6A3, AZGP1, CHI3L2, and IL36RA
Key Highlights:
-Fewer than 100 design attempts per target
-Binding, developability, and ~100 nM functional potency confirmed across 5+ -orthogonal assays
-Atomic accuracy confirmed by cryo-EM
Our approach addresses a critical challenge in therapeutic discovery: designing antibodies for "zero-prior'"epitopes, which we define as target sites lacking structural data from reported antibody-antigen or protein-protein complexes involving the target.
To our knowledge, this is the first demonstration of de novo design of full-length monoclonal antibodies (mAbs) against "zero-prior" epitopes with atomically accurate complex structures and functional activity.
