Bukti² menunjukkan bahwa ayam diperkenalkan ke Afrika melalui dua jalur sebaran utama: jalur Mediterania yang melewati Mesir sekitar tahun 300 SM, dan jaringan maritim Samudra Hindia pada milenium pertama Masehi.

Keberadaan garis maternal haplogroup D dari Asia Tenggara pada populasi ayam di pesisir timur Afrika dan Madagaskar merupakan salah satu bukti terkuat terkait penyebaran lintas samudra tersebut, hal ini mengindikasikan peran para pelaut penutur Austronesia, yang mengolonisasi Madagaskar sekitar 1500 tahun lalu, sebagai pembawa ayam yang memiliki garis keturunan maternal asal kepulauan Asia Tenggara.

Haplogroup D memiliki kaitan yang sangat erat dengan nenek moyang ayam di Asia Tenggara dan menunjukkan frekuensi tinggi pada populasi pesisir timur Afrika serta di Madagaskar.

Ayam dengan garis maternal haplogroup D tidak ditemukan di Semenanjung Arab dan Asia Barat; komposisi haplogroup di wilayah-wilayah tersebut didominasi oleh haplogroup E.

Di India, ayam dengan haplogroup E juga ditemukan dalam frekuensi tinggi (67%), sementara haplogroup D (22%) dan sisanya haplogroup yang lain (11%).

Di wilayah daratan Asia Tenggara (MSEA), seluruh haplogroup ditemukan (A hinggaa I), tetapi frekuensi haplogroup D dan E jauh lebih rendah dibandingkan haplogroup lainnya (masing-masing sebesar 4,8% dan 2,4%).

Sedangkan ayam di kepulauan Asia Tenggara dan Pasifik memiliki proporsi haplogroup D yang tinggi (84%), sesuai dengan sebaran penutur Austronesian.
