Mutually assured destruction wasn't an official doctrine. Rather, it was an acknowledgement of the reality of a war between two nuclear armed powers. And the key word there is "mutual." The Samson Option isn't mutual. Israel is the only nuclear armed power in that region. It is explicitly asymmetric.

MAD was basically "they won't bomb us because they know if they do, we'll launch ours and we'll all be dead."

The Samson Option is more "we will bomb literally anyone because if we go down, everyone goes down." It's explicitly a form of hostage negotiations, rather than a simple reality.
