I have not investigated any microbial biotransformations, so far I have been looking at the toxicity effects solely on the intestinal epithelial cells, however I do respect that gut microflora will play a part in drug metabolism!

Once our lab reopens, I will be performing these 5-FU studies on murine colonoid cultures to compare.
@DaveFernig @FHLSPGR @LivUniEngageHLS @LivUniISMIB Thank you @DaveFernig! I have not tested potential protective agents on enteroids, I have seen papers of in vivo work using protective agents against 5-FU, I would definitely be interested to try it in vitro! π
@LippyLiptrott@FHLSPGR @LivUniEngageHLS @LivUniISMIB@LippyLiptrott membrane and see if there are differences in the effects of toxicity. Comparison of the murine enteroids to e.g. human enteroid data has so far shown that murine enteroids are more sensitive to 5-FU doses.
@LippyLiptrott@FHLSPGR @LivUniEngageHLS @LivUniISMIB@LippyLiptrott Currently we are adding 5-FU to the enteroid media, meaning drugs are being exposed to the basolateral membrane of the epithelial cells. Colleagues have been working on microinjection techniques so we may be able to apply drugs to the lumen/apical (answer cont.)

@LippyLiptrott@FHLSPGR @LivUniEngageHLS @LivUniISMIB Hi @LippyLiptrott, I feel that the enteroids do reflect the in vivo environment well - we can see crypt and villus regions within the enteroids to reflect the same structures seen in vivo. I haven't personally investigated transporter expression in enteroids (answer cont.)
