Scientists just proved, for the first time in humans, that intermittent fasting activates autophagy - your body's cellular self-cleaning system.
A 2025 RCT published in the Journal of Physiology finally confirmed it works in living people.
Here's what the science says.
The study: 121 adults with obesity, randomised to 3 groups - standard care, calorie restriction, or intermittent fasting + time-restricted eating (iTRE) for 6 months. Researchers directly measured autophagic flux - a physiologically relevant marker in living humans. First time this had ever been done.
The result: at 6 months, the iTRE group showed a statistically significant increase in autophagic flux vs standard care (P=0.04). Calorie restriction alone? Not significant. It's not just eating less - it's the fasting itself that activates autophagy in humans.
What is autophagy? When you fast, insulin and glucose drop. mTOR - the cell's growth signal - shuts off. This activates the ULK1 complex, which initiates autophagy: degrading damaged proteins, dysfunctional organelles, and misfolded cellular junk. Nobel Prize-winning biology
A separate gene expression study found that 4 weeks of IF upregulated 3 key autophagy genes: LAMP2 by 4.2x, LC3B by 1.9x, and ATG5 by 1.4x - alongside significant drops in body weight, BMI, fat mass, and inflammatory markers. The genes themselves change in response to fasting.
How long do you need to fast? Current estimates from human marker studies: autophagy ramps up after roughly 14-16 hours of fasting. 16:8 (eating within an 8-hour window) appears sufficient to cross this threshold - you don't need 72-hour fasts to activate cellular clean up.
Why does this matter beyond weight?
