We then infect the bacteria with the phage and the old tail fiber gene will be switched with the new one in the viral genome by homologous recombination. The new virus then multiplies and assembles itself inside the bacteria. (3/3)
Last week we got our first significant result!
Thanks to our recombineering approach, we were able to swap parts of the T7 tailfiber for parts of the T3 tailfiber. Those new phages were then able to infect E. coli ECOR16, a strain that previously could not be killed by T7. (1/3)
Our recombineering approach uses the homologous recombination machinery of E. coli cells to insert a DNA sequence from a plasmid into the tail fiber gene in vivo. We first transform a plasmid containing the new T3 binding site codons of the tail fiber gene (red in the figure).
Last weekend, we participate in #Scientifica19, a fair organized by @ETH and @UZH_en to promote science to the broad public. To make our project understandable to our younger audience, we dedicated the lower part of our poster to them. They seemed to really like our phages!
The number of teams in Switzerland is creeping up, thank you @igem_uzh for organizing the first swiss meetup! We had a great time and really enjoyed hearing @EPFL_iGEM and @igem_uzh team projects and exchanging ideas! Can't wait to see you again in Boston!
Thanks to the support of @ArborBio and their myTXTL solution we were able to assemble T7 phage in vitro. We obtained 10^10 PFU/μL!! 🍾

Part of our team spent the weekend in Düsseldorf for the german meetup. It was a great opportunity for us to present our project for the first time to other iGEMers, get some feedback and discuss possible collaboration with other teams.
