Someone stole our female squid!
And swapped in a male.... unfortunately two males do not make for making squid babies!

12 hours to prep a sample and start collecting a developmental time lapse of a now 36 hour MBL squid embryo on the diSPIM light sheet microscope.
You can see how the two imaging arms collect images at -90 degrees to each other.

We got 4 new octopus yesterday - for an arm regeneration project that Kirsten Edepli is starting this summer.
This one pooped - not sure if it was shooting it me...! https://t.co/tH2J8ku3kj
I’m moving slow but getting better at running and even writing FIJI macros for processing diSPIM light sheet microscope data.
Here is the same data set from before of a 2 day old squid embryo with fluorescent dye labeling the nuclei - but now the two op… https://t.co/8QO1Wj1VYJ
I got basic diSPIM image processing worked out using FIJI - and generated a simple time lapse from the 15 hour squid embryo data set from Thursday!
The dye is fading from cell divisions and not sure if it’s laser or something else. Things will improve a… https://t.co/yO8chnSJFL
This has been a great week for lab milestones!
After a month and a half of YouTube training, Kirsten is running the diSPIM and we are getting our first developmental time lapse data sets.

Kirsten, Hari, Harsh, and I did what may be the first ever developmental time lapse of a cephalopod embryo on a light sheet microscope on Wednesday.
It was exciting but we used a 40x objective and because of the high magnification could only image a sma… https://t.co/ty6Crdllnp
Malaika continues her quest for Mnemiopsis ctenophores for single-cell sequencing by collaborators.
Tonight still zero - but she did find this gorgeous postlarval lobster in the plankton!

Pygmy squid Idiosepius pygmaeus vs MBL squid Doryteuthis pealeii
Hatchlings from today - Malaika is breaking jet propulsion in both - and testing if we can predict from the results of one at what viscosity the other one should break jet propulsion- base… https://t.co/34kE2aK3Lw

The hydrogel paper with Dirk Albrecht and Kyra Burnett is out!
It includes (I think) the first publication of live light sheet imaging in cephalopod - using pygmy squid hatchlings immobilized in hydrogel and fluorescent dye!

OceanX is posting some of our videos on Twitter for #cephalopodweek !
Friendly octopus hatchling is up - and Peter’s imaging of squid hatchling brains - cleared and labeled with dyes - is in the works!

It’s been an exciting weekend for live imaging - but I’m ending the night / morning with a massive set back.
All the gDNA that Kirsten and I have been preparing the last month is completely degraded due to a mistake I made.
We had resuspended the DNA … https://t.co/jHwtDU6jz3
