Our new paper in #PNAS is out - see here: https://t.co/x2Ct71Ghjz! Congrats to @ToniTraeger for leading such a work, uncovering fantastic insights into active site regulation in isomerases from their endogenous state :) A small thread to explain how disorder regulates catalysis:
We are excited to be opening 2 Ph.D. and 1 post-doc positions to work on cryo-EM and computational biology of plant protein complexes & their proteoforms (Projects A04, C04 and D01)!
Visit https://t.co/ocqy25c24N to be informed and apply ASAP! #SFB1664, headed by @quintlab
#KastritisLab strikes again: cell extracts and oxoglutarate dehydrogenase complex structure/function: Cryo-EM,cell biology,kinetics,XL-MS,Alphafold,HADDOCK: We visualise a gigantic 4 MDa active metabolon! paper is screened @BioRxiv; sneak-peak of main findings follow :-) (1/11)
(1/10) We combined #AlphaFold and #CryoEM to derive atomic models of MegaDalton protein community members from cell extracts! The efforts from the #KastritisLab together with @ktototak were extraordinary to reveal their architectures - see thread below:

(9/10) Our work shows the power of accurately modelling maps of intermediate resolution with #AlphaFold and #FindmySeq, revealing unique adaptations of native states - some required human intervention to resolve. We have a deep discussion on the impact of our work, please check!
(10/10) I would like to thank @BMBF_Bund for funding our research, @dfg_public for supporting my lab, @UniHalle, as well as @embl for the amazing collaboration!! And of course, @imjohnskal@Fotansky_16 @ctueting1 @hamdiphilum @ktototak for producing such interesting story!!
E3BP is not merely structural, and does not only bind the E3 - it is always in proximity of E2 monomers of the core, which are mostly in an open state configuration, showing functional implications of its positioning in the PDHc scaffold.

We then saw that in the native PDHc core, not only the E3BP, that binds the E3 is inside the core, but it is occupied by protein densities up to ~20%, showing that the scaffold is filled with polypeptide regions and is not empty.

We characterized down to atomic resolution the acetylation of coenzyme A in the giant, native PDHc metabolon from eukaryotic extracts! Congrats to @ctueting1 @Fotansky_16 @hamdiphilum @imjohnskal @JMuellerBoerger @SorokinaMarija @YasharSadian , see thread
