If I'm not mistaken, there were rumors that the main villain would be a swordswoman.
And through the set videos, it's possible to see a very prominent woman who people think will have a bigger role there, alongside the Hand. So, if the rumor is true, maybe the Hand really is the main antagonist (or final threat), or an eventual threat.
I think Scorpion, Tombstone, and the Hand will be planned threats, while Hulk will be an "accidental/eventual" fight, and there will probably be a fight against Sadie Sink's character, who is probably Jean Grey. I'm thinking that will be the final fight because the mystery is so great that I honestly think she'll end up being the main villain (since the trailer itself gave a lot of prominence to her character as a major threat, and she'll probably be the one to do something with the Banner device to get Hulk out).

Jean Grey will probably be the main villain (since she's the only one being kept completely secret, and the first trailer gave a lot of detail about her as a threat). Perhaps the final confrontation will be with the Hulk, because someone who correctly predicted the Punisher and Hulk's participation in the film said it would be Spider-Man and the Punisher against the Hulk, and I think he might be right that the Hulk will have a big confrontation there.

I can't find where I saw this now because I saw it in a random Twitter post, but practically I think it makes sense, since the spell only made everyone forget who Peter Parker is but doesn't completely undo Beck's speech; probably for them it only cut out the part where he says that Spider-Man is Peter Parker.
