(4/7) ScRNAseq revealed that the TIGIT-NECTIN2 L-R interaction in CD8 T cells and Macrophages was induced by chemo, and targeting this pathway in immune-competent patient-derived cultures showed T-cell activation.
(3/7) CD8 T cell interactions with M1 and M2 macrophages contributed to patient prognosis in Myelonets as well as at the tumor-stromal interface, where the interactions with M2 macrophages were mutually exclusive with CD8 T cell - tumor cell interactions.
(2/7) After analyzing >100 chemo-naive and chemo-exposed tumor samples, we identified stuctures called ”Myelonets” - networks of myeloid cells - recurring across patient samples and driving T-cell exhaustion.
