I am puzzled by results on EDS-SAG subcellular localization requirements. We did similar experiments [doi:10.1111/nph.18511] - NES may be confounding, but we did not see reduced function of E/S-NLS versions. Not the same species, but still very surprising.
it does not change deletion frequency, but point mutations increased by two-fold. Not new, TREX2 was used in several papers with @DanVoytas. But we put a number on it in https://t.co/r3ZrzBoKDA. by a direct comparison in stable transgenics. I hope this can be helpful for some.

We tested induction of chromosomal deletions. Apparently, frequency is higher when multiple sites flanking the fragment are targeted. Further, we tested TREX2 co-expression (with Cas9) in Arabidopsis...

@IzzySaur Hi, the Xanthos I worked with were very happy with NYG medium, solid or liquid. As an alternative to competent cells, triparental conjugation works fine. In most cases, there is no point in going liquid, they are more happy on plates.

A bit of a potpourri... we looked at PRR and TNL functions in Nbenth. Different from At, we do not detect a role of EDS1 complexes in Nb PRR signaling.
Always much criticism on peer reviewing, and I may also have been dissapointed before... but it really did a great job on this one. The reviewers proposed important experiments that cleared out last doubts on significance of the EDS1 triad in TNL-ETI.👍👏
