There are two major findings in this story.
2. Combining L-asparaginase with ASX-173, a newly developed ASNS inhibitor, significantly delays disease progression in ASNS-high ALL models in vivo, without prohibitive toxicity.
1. Despite initial efficacy of GCN2 inhibition in sensitizing acute lymphoblastic leukemia (ALL) cells to L-asparaginase treatment, a subset of Gcn2-deficient cells express ASNS at high levels in a Gcn2-independent manner, which eventually drives therapeutic resistance.
