Pregnancy involves dynamic interactions between maternal and fetal tissues that are difficult to capture in conventional in vitro models.

🤰 In this new publication, @akaLAPD, Sarah McColman, and colleagues review human pregnancy-associated organs-on-chips that model relevant systems, including the placental barrier and fetal membranes.

These models use engineered extracellular matrices and controlled flow to study biochemical gradients, mechanical forces, maternal–fetal transport of drugs, and more.

Key variables include maternal and fetal cell types, matrix composition and mechanics, and perfusion rate, all of which determine cell behavior, barrier function, and transport.

As the authors state, “When thoughtfully designed, pregnancy-associated bioengineered models can outperform animal models of pregnancy in terms of human physiological relevance and precise control of microenvironmental variables, and decreasing animal use can reduce financial and ethical constraints.”
