Announcing the first participant has been dosed in the Ph1 study for LRK-4189, a 1st-in-class targeted protein degrader of the lipid kinase PIP4K2C for the treatment of microsatellite stable (MSS) colorectal cancer (CRC). Learn more https://t.co/VkL1HNXG2l
We’re proud to present new data at #SITC2025. We will share preclinical research on our lead candidate, LRK-4189, which we anticipate advancing into a first-in-human clinical study soon.

LRK-4189: A Potential First-in-Class PIP4K2C Degrader Heads to the Clinic | https://t.co/0TG8TLqzom
PIP4K2C has minimal catalytic activity, but reported scaffolding roles make it well-suited for targeted degradation.
Building on insights from tool inhibitors and an academic degrader, Larkspur optimized a CRBN-recruiting molecule, LRK-4189, now planned for first-in-human study in MSS colorectal cancer.
Ex vivo CRC spheroid responses, supportive once-daily PK/PD, and xenograft proof-of-concept underpin the program.
