In this work, we provide a comprehensive overview of the emerging role of TIRAP as a critical signaling hub connecting pattern-recognition receptor activation with chronic inflammation, tumor progression, and therapeutic resistance across diverse cancers.

Toll/interleukin-1 receptor domain-containing adaptor protein (TIRAP) in Hepatic Stellate Cells: Beyond Membrane and Cytoplasmic Presence.
For years, we’ve known TIRAP (also called MAL) as a protein working strictly at the cell membrane to handle immune signals. But our team just found something completely unexpected: it actually moves into the nucleus during alcohol-induced liver damage.
What we found:
Moves to the nucleus:
Alcohol and LPS exposure activate TIRAP, leading to its translocation into the nucleus of hepatic stellate cells.
Activates HSCs:
Nuclear TIRAP promotes HSC activation and contributes to ALD progression.
Drives liver scarring:
Inhibition of TIRAP significantly reduces α-SMA and collagen, two key markers of hepatic fibrosis.

These findings highlight a promising new therapeutic target for chronic liver disease and fibrosis.
