The epigenetically enhanced EVs modified the transcriptional landscape of recipient hMSCs, promoting their mineralisation capacity. Epigenetic reprogramming provides a novel engineering strategy to enhance EVs regenerative potency as an acellular tool for bone repair.
Here we induced hyperacetylation via the histone deacetylase inhibitor TSA, augmenting osteoblast epigenetic functionality and promoting its mineralisation. EVs from epigenetically modified cells were enriched with pro-osteogenic microRNAs and transcriptional regulating proteins.
María Fernández-Rhodes from Midlands is explaining to us how to characterise and isolate extracellular vesicles. "We need to homogenate the research on how to isolate these vesicles"
