MSc Organic Chemistry | I read the peer-reviewed literature on NAD+, NMN & longevity compounds
Skeptical of supplement marketing. Grounded in mechanisms.
David Sinclair popularized taking 1g of NMN daily for longevity. But millions of people are blindly copying his stack and silently crashing their cellular energy. The missing piece? Methylation. A biochemical breakdown of why your NMN might be making you tired 🧵👇

If this made you rethink your stack, follow for one chemistry breakdown per day.
David Sinclair takes 1g of NMN daily.
Millions copy him.
Almost none of them know they're quietly burning through a methyl group reservoir their body can't easily replace.
A thread on the chemistry Sinclair doesn't talk about. 🧬👇
Signs your methyl pool is under strain:
Fatigue at doses that should work
Mood changes (serotonin synthesis is methylation-dependent)
Sleep disruption
Elevated homocysteine on bloodwork
These aren't NMN side effects. They're methylation depletion symptoms.
The ratio matters.
A rough biochemical guideline based on pathway stoichiometry:
500mg NMN → 500mg TMG minimum
1,000mg NMN → 1,000–1,500mg TMG
This isn't a marketing stack. It's a mechanistic requirement for SAM regeneration.
TMG (trimethylglycine) donates methyl groups directly via the BHMT pathway, bypassing the folate cycle.
It remethylates homocysteine back to methionine.
Methionine → SAM → the cycle restarts without the SAH accumulation bottleneck.
SAH → homocysteine via hydrolysis.
Elevated homocysteine is not a minor footnote.
It's an independent cardiovascular risk marker (PMID: 15767285) and a correlate of cognitive decline (PMID: 11925300).
This is the biochemical cost of running the salvage pathway without methyl support.
When you flood the pathway with NMN, you accelerate NAD+ synthesis.
But you're also cycling SAM faster.
The byproduct is S-adenosylhomocysteine (SAH) — which accumulates, inhibits methyltransferases, and blocks the very reactions that regulate gene expression and neurotransmitter synthesis.
NMN works by raising NAD+.
But the Nampt-driven salvage pathway — the primary route NMN takes inside your cells — consumes S-adenosylmethionine (SAM) at every cycle.
SAM is your body's universal methyl donor. It runs over 200 methylation reactions.
I skipped the wellness blog fluff and ran a rigorous, literature-backed biochemical audit on NMN—breaking down the exact dosing protocols, safety data, and bioavailability mechanisms you need to know.

Most "anti-aging" advice is pure marketing hype. But as an organic chemist, there’s one molecule that actually commands serious attention in the lab: NMN.
Here is the unfiltered cellular reality of how Nicotinamide Mononucleotide impacts your biological clock. 👇
But here is where the industry tricks you: Not all NMN is created equal.
From delivery mechanisms (sublingual vs. liposomal) to raw purity standards, the biochemical matrix determines how much actually survives your digestive tract to reach peripheral tissues. Most retail options are a waste of capital.
Enter NMN (Nicotinamide Mononucleotide).
NMN acts as a direct, rate-limiting precursor. Once absorbed, your body rapidly synthesizes it into functional NAD+, effectively re-charging your cellular batteries and fueling the pathways that mitigate biological decay.
The core problem: As we age, our NAD+ levels collapse by up to 50%.
Think of NAD+ as the cellular currency required for DNA repair, sirtuin activation, and mitochondrial energy. Without it, your metabolic engine stalls. But you can't just swallow raw NAD+—the molecule is too large to cross cell membranes efficiently.
Anthropic just dropped a 31-page prompting guide.
Here's everything you actually need (in 10 rules):
1. You write "review this contract" and pray.
Fix: Name every output. "Review this contract. Flag risks per clause. Rate severity 1-5. Return as a table."
2: You say "summarize this" on a 40-page report.
Fix: 4.8 sizes the answer to the input. Cap it: "5 bullets. Each under 15 words. Start each with an action verb."
3: You write "don't use jargon. don't be salesy."
Fix: Negative instructions don't stick.
Flip them: "Write in plain English a 16-year-old could read aloud."
4: You type "can you help me with the email?"
Fix: Each verb ships something. For example: "Go to Gmail. Find [contact]. Write the send-ready reply. Under 90 words. Tone: confident, casual."
5: You wait for Claude to web search on its own.
Fix: Claude opus 4.8 calls fewer tools than 4.6.
Force it: "Use web search aggressively. Verify every claim with at least 2 sources."
6: You miss the warm tone from old Claude.
Fix: Claude opus 4.8 is direct. Almost zero emojis. Paste 2-3 sentences in the voice you want.
Tell Claude to match the rhythm.
7: You ask for "a landing page" & get bare minimum.
Fix: Drop this one line on every creative task
→ "Go beyond the basics."
It's from Anthropic's own doc.
8: You forget Claude 4.8 doesn't reason by default.
Fix: They call it "Thinking." Effort: High
Add this at the end: "Think before answering (maximum reasoning)." Free upgrade. Every time.
9: You rewrite the same prompt 14 times a week.
Fix: A skill is a command with instructions pre-built.
Write the same prompt twice? Make it a skill.
10: You assume Claude knows what you meant.
Fix: Old Claude 4.6 guessed.
New Claude 4.8 does exactly what you typed.
Spell it out. Output. Order. Length. Tone. Format.
If you don't say it, you don't get it.
