Take-home message: Xist deletion in the B cells of female mice can promote lupus-associated immune abnormalities, supporting a role for XCI in protecting against autoimmunity. Sometimes the genes you express are just as important as the ones you silence.

One signaling pathway downstream of T cell receptor activation is the canonical NF-kB pathway. We discovered that chemical inhibition or genetic deletions that block NF-kB signaling prevents Xist RNA ‘clouds’ in mouse and human T cells, and altered X-linked gene expression!
