@DrKlemow Synechococcus is a cyanobacterium that can be found throughout various aquatic environments, they are not just limited to lakes in upstate NY.
@DrZ_Micro I think there was more pressure to lose the plasmid in the stationary phase because the population of cells had reached optimum growth. Any strains that were not carrying the plasmid did not have to expend the extra energy associated replicating and expressing.
@GossardKelsey One possible factor affecting microcolony formation is the vessel the cultures are growing in, due to differences produced in oscillation of the media when in the shaker.
@GossardKelsey Yes, in flask 3 less microcolony formation was observed and the resulting RFU readings were higher than the other replicates. All four replicates were identical and the cause of microcolony formation is unknown.
