After more repeats, we can't reproduce this anymore unfortunately. In fact we are now seeing that BX795 reduces transduction efficiencies and viability in some cell lines.

@NikoMcCarty@iGEM Add a furin cut site + GSG before the 2A to reduce extra amino acids on your N terminus and boost cleavage. Also, plasmids may replicate in mammalian cells using ebna-1/oriP or S/MAR (not sv40 ori / 293tt, it's transient). We have seen oriP work and we haven't tried S/MAR.
