Excited to share another recent paper from the lab, in collaboration with @lucapinello and @danielevanbauer. We leveraged allele distributions after dense editing to identify the regulatory potential of individual nucleotides within #enhancers:

A HNF1A-A1CF transcription-splicing axis is suppressed in β cells from #T2D individuals, while genetic variants reducing pancreatic islet A1CF are associated with increased glycemia and T2D susceptibility https://t.co/jPa3vY8gY5
🚨Paper alert!!🚨
I am proud to announce that my postdoc work is now published in @Cell_Metabolism.

Glucose tracing experiments show increased incorporation of carbons into TCA-cycle intermediates, further highlighting the importance of mitochondrial metabolism for the increased functionality. 4/5
We have previously reported SC-islets to have aberrant reactivity to pyruvate in vitro, which we now show is resolved upon implantation. A promising result as the maintenance of pyruvate reactive insulin secretion could lead to exercise induced hypoglycaemia. 5/5
Why does it take SC-islets over 2-months to humanise mouse blood glucose when human islets achieve this feat a few weeks? This could be due to changes occurring to SC-islets upon engraftment. 1/5

Really excited to share that I got an #ERCCoG from @ERC_Research The project EARLYFATE will explore the interplay between DNA damage responses and the very first events of mammalian development. Huge thanks to all colleagues, former mentors and lab members for their support!
We had a great 5th IPITA/HSCI/Breakthrough T1D meeting on Stem Cell-Derived Islets recently at Harvard Medical School! I am very excited about the direction this field is taking to develop cell replacement therapy for people living with #T1D.
Save the dates! June 11-13, 2025.
The European Islet Study Group Meeting of 2025 will take place in Malmö, Sweden hosted by our friends at Lund University Diabetes Centre.
