Today, I turned the big 5-0 & grateful for this incredible day I had filled with love, laughter, messages, calls & so much warmth from family & friends.

Yet, as always on special days like this, there was an ache in my heart. I missed my sweet Ashley more than words can say. She should have been here blowing out candles with me, teasing me about getting “old”, & filling the room with her beautiful smile & that contagious joy. I know she’s celebrating with me in her own perfect way, but oh, how I wish I could hug her one more time. Life is a mix of profound gratitude & quiet longing. Today held both & that’s okay.

Thank you again for making my 50th so meaningful. Here’s to the next chapter, carrying Ashley’s light with me every step of the way.

When my precious 13 year old Daughter, Ashley, passed away from brain cancer, I chose not to let grief consume me. Instead, I channeled that profound pain & heartbreak into founding Cancer Response Team (@Nonprofit4Kids), a nonprofit dedicated to supporting Children battling cancer & their families.

Through the deepest sorrow, I persevered to create a lasting legacy for my Ashley, one that honors her courage & ensures her experience helps ease the journey for others facing this devastating fear.

Charlotte has a plan. She is determined that in six weeks’ time  by her birthday at the latest  she will be walking again.

Looking back now, ten weeks from this moment she would be nearly gone.

At present, her metabolic rate is extremely low, which explains the significant weight gain. It is something that depresses her deeply, affecting not just her body but her spirit. She wants her independence back, her mobility, her sense of self  and she clings to that hope with everything she has.

In early 2024, researchers at Massachusetts General Hospital reported preliminary findings from a small phase 1 clinical study (the INCIPIENT trial) of an experimental CAR T-cell therapy for recurrent #Glioblastoma The approach was developed in collaboration with Harvard Medical School & is known as CARv3-TEAM-E.

This next generation therapy was designed to help immune cells recognize multiple tumor markers at once, specifically targeting the EGFRvIII tumor specific antigen via the CAR while secreting T-cell-engaging antibody molecules (TEAMs) against wild type EGFR addressing some of the ways glioblastoma can evade treatment due to tumor heterogeneity. In the first three individuals treated (between March and July 2023), researchers observed rapid & dramatic tumor shrinkage on MRI imaging shortly after a single intraventricular infusion.

One participant showed near complete regression within days (confirmed over the next two weeks), while another experienced substantial reduction starting on day 2 (progressing to ~60% by day 69 & sustained beyond 150 days at last assessment). The third had rapid initial shrinkage that was transient.

Scientists emphasize that these are very early, proof of concept results from just a few patients in a safety focused trial, with no dose limiting toxicities but some manageable grade 3 side effects like encephalopathy.

Follow up data presented at the 2025 ASCO Annual Meeting (from seven patients receiving up to six doses after lymphodepleting chemotherapy) showed stable disease in five, including one with a 33% tumor reduction after two infusions, though responses remained transient overall due to factors like reduced T-cell persistence & anti-drug antibodies.

Much more research is needed to understand long term outcomes, safety with repeated dosing & broader effectiveness.
