What is a professor actually looking for in your PhD cover letter? When I read one, I’m usually trying to answer four questions very quickly.
Why me?
Not “I am interested in your research.” Show me that you know what my lab is currently working on. One recent paper, one research problem, one specific connection is usually enough.
Why you?
Don’t give me your entire CV. Show me the 1 or 2 experiences that make you useful for this project. If your experience is in analysing longitudinal clinical data, tell me that when my project requires exactly that. Make the transferability obvious.
Why this research question?
This is where I look for evidence that you’ve actually thought about the science. What did your previous work make you curious about? What limitation did you encounter? What relationship or assumption would you like to investigate further?
Why should I believe you can do it?
This comes from specifics. A methodological decision you made. A problem you solved. An unexpected result you interpreted. A technique you didn’t just learn, but actually used to answer a research question.
By the end, I should be able to summarise your letter in one sentence:
“This person has worked on X, understands Y, and could contribute to Z in my lab.”
If I can’t do that, the letter probably needs restructuring, regardless of how polished the writing is.
That’s also how I review cover letters with my mentees. We strip away the generic language and rebuild the letter around what the PI needs to understand about you within the first minute.

One of the biggest mistakes I see applicants make is finding their dream fully-funded Master's or PhD, only to miss the deadline because they are scrambling for paperwork.

How to read a PhD vacancy backwards to understand what the PI actually needs. Tips for CV and Cover Letter⬇️
Look closely at the verbs. “Develop and validate” suggests they need someone who can build, test and troubleshoot. “Characterise and optimise” points towards experimental judgement and iteration. “Analyse and model” tells you they need more than familiarity with a software package. The wording is often a better indicator of the actual role than the generic “requirements” section.
Then take the research objectives and reverse-engineer the capabilities behind them. If the project says, “Develop a machine-learning model to predict treatment response using multimodal clinical data,” don’t respond with “Python, machine learning, data analysis” on your CV. Surface evidence: “Developed and validated a predictive model using 12,000 patient records, compared three algorithms and addressed missing and imbalanced data.” Now the PI can see that you’ve already handled pieces of the problem they are hiring for.
The same change should happen in your cover letter. Don’t write, “I am excited about this position because I have experience in machine learning and clinical research.” Translate your experience into their research need: “Your project requires integrating heterogeneous clinical datasets into a validated predictive framework. In my Master’s research, I worked with longitudinal patient data and compared multiple modelling approaches while troubleshooting missing and imbalanced data.” The second version gives the PI a reason to believe you can contribute.
I also look for requirements that appear repeatedly across the vacancy. If the description keeps mentioning independent research, troubleshooting, quantitative analysis and collaboration, I treat those as selection signals, not filler. Your CV should contain evidence for each. Your cover letter should connect the strongest evidence to the project. Your interview examples should then prove that you actually did those things.
Before applying, I would literally make three columns: What the PI needs → Evidence I have → Where I show it. If the evidence exists but is buried in your CV, you have a positioning problem. If you have the skill but no evidence, you have a credibility problem. If you have neither, you have a genuine application gap that needs addressing.
This is one of the things I do when I work with applicants on their CV, cover letter and PhD shortlist. I don’t just make the documents sound better. I work backwards from the research position to decide what the PI needs to see, which parts of your experience actually prove it, and what should be removed because it adds noise.

How does a professor decide whether a PhD applicant is actually ready for research. Here’s what they look beyond your GPA, university rankings, or the number of papers on the CV.
1. Can you explain why you made a research decision?
They want to know why you chose that method, changed an experiment, rejected an approach, or followed one result further.
“I used Python for analysis” is a skill.
“The initial model was overfitting, so I compared two alternatives and changed the validation strategy” is research judgement.
2. What happens when the project doesn’t work?
This tells them a lot.
Did you troubleshoot systematically? Change the hypothesis? Check whether the problem was experimental or analytical? Decide when an approach wasn’t worth pursuing?
A failed experiment can be stronger evidence of research readiness than a successful experiment you simply followed a protocol for.
3. Can you identify what your own work couldn’t answer?
After every project, they want to know:
What did you establish?
What remains uncertain?
What assumption did you make?
What would you test next?
If you can answer those questions, you’re already thinking beyond the assignment.
4. Do you understand your contribution?
Being part of a research project isn’t the same as owning a piece of it.
Theh look for applicants who can tell me exactly what they contributed, what decisions they made, and what changed because of their work.
5. Can you connect your previous work to a new problem?
This is particularly important when your background doesn’t perfectly match the PhD.
You don’t need to have already done the exact research.
You need to show that your methods, reasoning, or previous questions can transfer to it.

A limitation tells you what the authors could not do. A research gap tells you what still needs to be understood.
That distinction matters when you’re writing to a potential supervisor.
One method I use is an argument map.
Take the paper and reduce it to:
Claim → Evidence → Assumption → Weakness → Unanswered question
For example:
Claim: X predicts treatment response.
Evidence: The authors tested X across 3 patient cohorts.
Assumption: The relationship remains stable across populations.
Weakness: All three cohorts came from similar demographic backgrounds.
Question: Does the relationship hold in a more diverse population?
Now go one level deeper.
Look at what the paper didn’t have to test because the authors made an assumption.
Ask:
* What variable was held constant?
* What relationship was treated as established?
* What population was excluded?
* What alternative explanation wasn’t tested?
* What happens if the method is applied in a different context?
* Which result is being interpreted more confidently than the evidence allows?
Then map the lab’s last 3 to 5 papers together.
If the same assumption keeps appearing, you’ve probably found something more valuable than a sentence buried under “Future Work.”
You’ve found a research direction.
That’s what you can bring into your cover letter:
“Across your recent work on X, Y and Z, I noticed that ___ remains largely unexplored. My experience with ___ could allow me to investigate whether ___.”
Now you’re not writing:
“I am interested in your research.”
You’re showing the professor what you’ve understood, where the question could move next, and why you might be useful in answering it.

PhD Cold email checklist (what professors actually look for)
1. Purpose of the email
* Why this lab and why now?
* One research question/direction only. Don’t mention three interests.
* Don’t start with your background. Start with the scientific problem.
* Within 20 seconds, they should know why you’ve emailed THEM.
2. Connection to the lab’s work
* Read 3–5 recent papers, not just the most cited one.
* Don’t summarise findings.
* Identify one recurring limitation, assumption, or future direction.
* Connect it to one of your projects.
* Show where your work overlaps. Don’t make them figure it out.
* End with one possible extension or question. Doesn’t have to be groundbreaking. It just has to show you’ve thought about the problem.
3. Call to action
* Don’t ask, “Are there any openings?”
* Ask whether your background could contribute to their ongoing work.
* Mention CV attached.
* If applicable, attach a one-page research brief.
* Keep the ending natural. You’re opening a conversation, not asking for permission.
