Just published in Nature (June 2025), Andersen et al. (@EAChiocca, Reardon, @QuintanaLabHMS) developed a novel viral barcode interaction-tracing (RABID-seq) platform, uncovering previously unknown immunosuppressive interactions between astrocytes and glioblastoma (GBM) cells.
Through a bidirectional ANXA1–FPR1 signaling axis, ANXA1 expression in astrocytes suppressed NF-κB inflammasome activation, thereby driving immunosuppression, while FPR1 activation in GBM cells inhibited immunogenic necroptosis. Targeting this axis resulted in enhanced infiltration and activation of dendritic cells (DCs), tumor-specific CD8+ T cells, and macrophages, significantly augmenting antitumor immune responses.
The RABID-seq method developed here, if integrated with spatial analyses, can offer unprecedented resolution into the complex cell-cell interactions within the GBM TME. Critically, identification of the astrocyte–GBM ANXA1–FPR1 axis as a key mechanism of immune evasion and tumor progression provides a promising target for therapeutic intervention, either as monotherapy or as part of combination strategies to significantly enhance the efficacy of immunotherapies for GBM.
