This is a list of the best longevity compounds in the public DrugAge database. The list is sorted by maximum lifespan for male mice. It's a little bit sad that the database only contains data to November 2021. It seems also missing some data like the data for females for the compound 17-a-estradiol etc. Would be awesome if we could get an updated database soon because I know many people in the longevity field use it.
When I look at the list there are many compounds that I have never heard of before. There are also lots of data gaps in multiple species on almost all compounds. Rapamycin has the best data coverage.
The compounds that I think have interesting lifespan data are Aspirin, NAC and of course Rapamycin. Here is a very simplified explanation of these three.
Aspirin = a medication for reducing pain, fever and inflammation. FDA approved for lowering risk of heart attack and stroke.
NAC = an amino acid supplement for raising antioxidant levels and reducing oxidative stress and inflammation. It is also a FDA approved medication for treatment of certain diseases.
Rapamycin = a FDA approved medication for organ transplantation to suppress the inflammatory response by the immune system so that it does not reject the new organ. Another main area it is used in is cancer treatment.
An interesting insight when I wrote the explanation above was that the common pattern between the three compounds is that they reduce inflammation. This reminds me of the interview I had with the Rapamycin researcher Matt Kaeberlein in which he talked about the aging immune system and how Rapamycin may improve it.
