We show that the bacterial SMC Wadjet complex cleaves plasmid DNA through loop extrusion.

Combining in vivo and in vitro analysis, we find that Smc5/6 preferentially loads and extrudes loops of transcription-induced positive supercoils, thus promotes local cis-interactions.
