Sweet moment in life, our 25th lab anniversary. In a circle, we looked at each other, a group of people with shared values and a soft approach, who want each other to succeed and be happy. (1/2)
Had the chance to hear Chris Voigt present this work at a SynBio conference a few months back- absolutely jaw-dropping science. Huge congratulations to the team on this incredible project!
> bacteria were engineered to make molecules that absorb light in unique ways.
> the microbes were buried in soil.
> using a drone with a hyperspectral camera, one can "see" where the microbes are buried from ~300 feet away.
one of the most sci-fi papers I've seen in awhile.
I am putting some effort in keeping track of the conferences on Theoretical Biology and Quantitative Life Science. If you like to add some, please check this page. Help the community!

Really cool!
1st thought - So basically we can have "multi-copy plasmid" by transforming a multimeric plasmid with BAC origin?
2nd thought - Jeez. How many clones in a combinatorial library may just be hetero-multimers?!! if they cannot be resolved later in recA strains...
@HaotianGuo_qb 2. Regarding libraries, it depends on the strain they are built in. Cloning strains are surely a safer way to go. In any case (hetero)multimers are not necessarily inherited. In any case, double-checking hits after any library screen would be beneficial: first, by sequencing the
Understanding multimer formation and using the MG1655 ΔrecA strain can significantly improve the design and reliability of synthetic biology experiments. Last but not least, shoutout to the team: David Glass,

Preventing plasmid multimer formation in commonly used synthetic biology plasmids
We survey and present a new way to quantify a hidden threat to synthetic biology - plasmid multimers. See the 🧵 for a tweetorial: 1/7
Deleting recombinases banishes multimers. The traditional cloning strain JM109 and a modified MG1655 ΔrecA strain prevent multimer formation. These are valuable tools for synthetic biologists to ensure reliable plasmid-based circuits. 6/7
