What is the relationship between #rapamycin and #autophagocytosis or #autophagy in the #fruitfly #Drosophila #melanogaster (Drosophilidae, #Brachycera; photo 1)? And how do these #interrelations affect the consequences of global warming and dry seasons with increased temperatures in #insects ?
Since weather records began, global surface temperatures have increased by an average of 1.15-2.20°C. The consequences of #globalwarming are clearly noticeable worldwide: warmer summers, persistent #drought periods and forest fires represent a #challenge for all organismic life. Individuals of various species die or their normal biological processes are hindered by #heatstress. Homoiothermic animals have to use energy to regulate their body temperature when they are exposed to excessive heat. Cold-blooded animals such as #arthropods or reptiles must seek cooler hiding places or have special #adaptations to prevent their bodies from overheating.
Using the model organism Drosophila melanogaster, researchers Q. Willot et al. (2023) examined the connection between heat tolerance and autophagy. They concluded that in fly individuals fed with the substance rapamycin, the rate of autophagy increased, which led to a noticeable increase in heat tolerance in experiments, in which the animals were exposed to higher temperatures. Heat stress reactions in the form of falling down with immobility occurred distinctly later.
Autophagy is an important mechanism at the #cellularlevel that leads to the efficient breakdown of excess #proteins and outdated cell #organelles. This increases vitality and adaptability of the entire organism. There is also a connection between autophagy and aging processes in mammals: as a vertebrate ages, the autophagy rate decreases.
How does autophagy work in general? In #macroautophagy, a membrane section of the endoplasmic reticulum (#ER) surrounds the molecules to be broken down and then migrates in the form of a double-membrane #autophagosome to a #lysosome, which finally breaks down the contents using hydrolases (Photo 2).
During microautophagy, the substances to be broken down are absorbed and decomposed directly by the lysosome (Photo 2).
Rapamycin is a macrocyclic lactone produced by the bacterial species #Streptomyces #hygroscopicus (Streptomycetaceae, #Actinobacteria). The lactone is known to produce an #immunosuppressive effect by binding to a specific protein (intracellular cytosolic protein), which combines with an enzyme (serine/threonine protein kinase) that inhibits the protein.
In mammalian experiments, rapamycin had a general life-prolonging and dementia-reducing effect, also due to the suppression of transcription/translation of various genes.
According to the authors mentioned above, rapamycin has an effect on insects that they studied on fruit flies, in the form of increased #heattolerance. What does this tell us?
The authors' findings indicate which mechanisms can trigger increased heat resistance in insects, namely the initiation of reactions that lead to #selfcleaning (autophagy) within a cell. In the case of flies fed with rapamycin, the target of rapamycin (#TOR) is inhibited, whereby autophagy is initiated in local areas of the fly #gut. Author A.
