Overall we are confident that #METPlatform could help to identify effective drugs against metastases and even provide a clinically-compatible strategy to expand the benefits of personalized medicine to systemic disease.
Omics: #METPlatform helps to understand why metastases respond to specific drugs, identifies biomarkers and strategies that could be exploited to target resistance before it happens. We report an interesting combination with a drug currently used to treat psychiatric disorders.
A patient “avatar”: #METPlatform might be even used to predict the response of patients to a drug in real life. We demonstrate that responding/ non-responding PDOC to the standard of care in glioblastoma correlates with clinically-established biomarkers.
Compatible with patients: #METPlatform is fully compatible with patient tissue. Patient-Derived Organotypic Cultures (PDOC) were derived from neurosurgeries of brain metastases where one of the top hits from the screen was validated.
Predictive value: #METPlatform prioritizes inhibitors that work in vivo, including a novel model of local relapse after neurosurgery, which has direct clinical implications as well as opens unexplored biology. Exciting!
Versatile/ Flexible: #METPlatform prioritizes different compounds than traditional in vitro platforms and allows to define drugs effective for preventing and/ or for treating metastasis, including potential drugs against the microenvironment.
All organs: #METPlatform is compatible with multiple organs affected by metastases. We applied it to challenge the viability of metastases growing in the brain, an unmet clinical need partly because of exclusion of patients from clinical trials
How to prioritize drugs that work in organs affected with #metastases without the need of using in vivo approaches? We report for the scientific community #METPlatform, an ex vivo #drug_screening platform using organotypic cultures (thus including the #microenvironment).
