As #PHAGEATAC uses fixed cells rather than nuclei for #scATACseq, it is able to leverage the recently described acquisition of mtDNA fragments for mtDNA-based genotyping and clonal tracing via mtscATAC-seq, introduced by @LeifLudwig and @CalebLareau.

We generated phage nanobodies against multiple immune cell surface markers and benchmark our #PHAGEATAC results against available #CITEseq data on human primary cells.

Instead of barcode-conjugated TotalSeq antibodies, #PHAGEATAC leverages phage display and uses nanobody-displaying phages for single-cell multiplex protein detection. The hypervariable nanobody CDR3 within each phagemid acts as a unique genetic barcode for epitope detection.
