In the study "Extracellular Matrix Mechanical Cues (Dys)regulate Metabolic Redox Homeostasis Due to Impaired Autophagic Flux" by Heloísa Gerardo et al., #Resipher technology was employed to measure oxygen consumption rates (OCR) in mouse hippocampal neuronal (HT22) cells under varying extracellular matrix (ECM) stiffness conditions.
The study revealed that prolonged exposure to soft ECM conditions (1–2.5 kPa) disrupted mitochondrial bioenergetics, reduced redox homeostasis, and impaired autophagic flux in HT22 cells. SH-SY5Y cells and primary mouse neurons were also used to validate the broader effects of soft ECM on neuronal metabolism, mimicking aspects of neurodegenerative disease pathology and offering a framework for improved in vitro disease modeling. We are excited about this recent publication by Cloé Tessier et al., where the researchers developed a protocol for real-time measurement of mitochondrial bioenergetics in 3D-cultured brain tumor stem cells (BTSCs) using the #Resipher system. Given the critical role of mitochondria in cancer metabolism, this protocol enables precise, high-resolution monitoring of oxygen consumption rates (OCR) in patient-derived BTSC spheroids.
The study outlines key optimizations for cell density, mitochondrial respiration modulator concentrations, and real-time analysis using Resipher, a device that allows continuous, non-invasive metabolic assessment in standard culture conditions. This protocol advances our ability to study mitochondrial dysfunction and bioenergetics in 3D tumor models, paving the way for improved metabolic-targeting strategies in cancer research.
