But it also takes place in cancer! We next screened a collection of 500 cancer cell lines from the PRISM collection @broadinstitute and found high activity in certain cancers, such as #melanoma and #glioma, with a single common denominator: #UPP1! #CancerMetabolism 8/10

This story started when we performed an #ORFeome over-expression screen to find genes involved in surviving #glucose deprivation. To our surprise, the main hits were #UPP1 and #UPP2: two #uridine #phosphorylases with no prior link to energy metabolism. 2/10

What cells can naturally do that? We screened the #PRISM 482 cancer cell line collection, comparing growth on uridine vs glucose and found that lots of cancers can grow on uridine, in particular #melanoma via #MITF. All have in common one thing: high #UPP1! (3/9)

It started when we screened an #ORF library for genes that promote growth in no glucose and found two hits: #UPP1 and #UPP2. Both supports growth on reformulated media with uridine REPLACING glucose, and UPP1-expressing cells grow *as fast* on uridine as on glucose! (2/9)
