This "overstimulation" chronic activation leads to exhausted Tcells. Why mechanisms that reduce the T-cell stimulation or interrupt can be so powerful to reverse T-cell exhaustion. #ITK integrates #TCR signal and #CD28 co-stimulation, but if you inhibit ITK you dampen signal and lead to increased tumor fighting and stem-like self-renewing CD8+ Tcells that have memory like capability for strong anti-tumour response.
$CRVS ITK inhibitor begins Ph1b/2 solid tumour trial in next few months to show this powerful effect and synergy with anti-PD1 therapy.

We know $CRVS oral #ITK inhibitor #soquelitinib MOA reduces strength of TCR/CD28 co-stimulation and this paper’s conclusions support Tregs are being made by ITKi including #epigenetic changes in #TSDR i.e., reduced demethylation and increased acetylation which I previously discussed which make #FOXP3+ persistent and results in more suppressive Tregs for treating #autoimmune disesases, i.e. #atopicdermatitis, #foodallergies, #asthma, etc.
Explains long-duration benefit $CRVS is seeing in atopic dermatitis study with disease control still occurring 60 days after treatment stopped. Looking forward to hearing more on this novel mechanism and Treg effect on May 7, 2025 in San Diego.
