We acquired a large-scale mixed-species benchmark, with variable background, to comprehensively assess quantitative accuracy of proteomics.
Key features:
- 192 runs, 0.75ng - 15ng of variable human cell line backgroud.
- Can dissect the impact of both random and systematic errors.
- Can test how quantitation algorithms scale with experiment size and sample heterogeneity.

Data from the second human treated with a prime edited medicine: a 56-year-old male with chronic granulotamous disease (CGD) had his bone marrow prime edited to insert the missing GT in NCF1, restoring NADPH oxidase function in neutrophils (DHR positivity) several fold beyond the therapeutic threshold. His intestinal inflammation is now normal, and his prime-edited HSCs rapidly engrafted with 15 days post-transplant, consistent with well-tolerated editing.
Each new therapeutic technology is dependent on brave patients willing to be among the first humans to receive a new type of treatment. For prime editing, that's this patient, and an 18-year old who also responded in a similarly encouraging manner.
Patient #2 sent our lab a kind message, noting that helping other patients with rare diseases was deeply meaningful to him.

"Development and extensive sequencing of a broadly-consented Genome in a Bottle matched tumor-normal pair" https://t.co/lfEODD1s1E congrats to the @WeillCornell lab team, Justin Zook, and @NIST for this key resource!
The authors also use TAP(https://t.co/wvtmkvMjW9) to evaluate the developability of the generated sequence in silico, and find that the vast majority pass the developability threshold compared to clinical Abs.

We retrospectively analyzed the reproducibility of 400 papers in the field of #Drosophila immunity. Irreproducibility was rather low. High-impact journals, top institutions and PI training influence replicability levels. #Reproducibility#LifeSciences
