Glycine is a powerful tool against kidney stones, suppressing crystal formation and promoting oxalate excretion.

Researchers examined human urine from people with kidney stone disease,

and they also did experiments in animals who were given ethylene glycol, which causes kidney stones by metabolizing into oxalate.

Kidney stones are often made up of crystals of calcium oxalate.

They first showed that people who had calcium oxalate kidney stones also had less glycine in their urine.

What was incredible was that glycine treatment almost completely prevented kidney stone formation.

After giving the animals ethylene glycol, the animals who also got glycine had:

➟ Almost zero crystal deposition
➟ Lower oxalate excretion
➟ Higher citrate excretion (binds calcium, preventing kidney stones)
➟ Higher urinary pH (acidic urine drives stone formation)
➟ Increased glycine excretionHow was glycine able to do this?

One of the key mechanisms seems to be that it decreases the expression of an oxalate transporter - Slc26a6.

This transporter takes oxalate from renal tubule cells and puts it into the urine.

Less oxalate in the urine = less opportunity for stone formation.

Glycine also had an impact on the citrate transporter.

Here it DECREASED the citrate transporter Nadc-1.

This transporter takes citrate from the urine and brings it into kidney cells.

Less of this transporter means more citrate in the urine, which binds to calcium and prevents kidney stones. Since glycine did not impact the gene expression of Nadc-1,

the researchers looked at if there was a way it could possibly prevent the existing RNA from being translated into protein.

Glycine increased the levels of this micro RNA (miR-411-3p).

Micro RNAs are genetic material that don't code for genes but instead impact gene translation - how they get turned into proteins.

miR-411-3p was able to suppress the amounts of both transporters.

This is how glycine prevented the kidney stones.

Human equivalent dose was roughly 2-3g of glycine per day.
