All other Joestar were alive at the same time DIO was, since DIO possessed Jonathan body, Jonathan’s body send a signal to all the bloodline descendants *forcing* their stands to awaken (hence the plot part of Part 3 trying to save Holly).
Stands are always there, as stated to be part of one’s soul, that’s why Stand’s abilities are linked to one’s master abilities (Joseph’s hamon, Kenzou’s Feng-Shui) and this is why non-Joestar characters like Avdol or Polnareff had their stands. Jolyne didn’t “needed” to get arrow-hit, that was just a way to force the awakening.
