Our paper is out today in Cell, "Trans-regulatory gene mapping prioritizes disease drivers in asthma", https://t.co/EcuxU2XIO3 🎉 GWAS hands us thousands of disease-linked variants, but the genes that actually drive the disease often aren't the ones sitting at those variants.
For asthma it flagged 21 DPGs, many missed by standard methods (GWAS, burden tests, PoPS, TWAS). In CRISPR screens in lung epithelial cells and T cells, most led to changes in epithelial integrity or IL-13 production, a far higher hit rate than for other genes (e.g., GWAS genes)
We first tested it where the biology is already well understood: 27 blood traits. DANDELION's genes were enriched up to ~22x in known Mendelian blood-disorder genes, a sign it's finding real biology. Reassured, we turned it on a disease we understand far less: asthma.
So we built a method we call DANDELION 🌱 It follows the trail from a GWAS variant, to the gene it's quietly regulating from far away ("in trans"), to the disease. Combining that with rare-variant data, it spots close-to-disease genes other methods walk right past.
Those variants usually act from a distance, tuning other genes elsewhere in the network. The genes we really want are the ones closest to the disease risk itself, where the interesting biology (and maybe future therapies) tends to be. Those have been hard to find.
