Since mold cultures may take 1-4 weeks of incubation and requires highly skilled microscopic identification, ITS NGS can identify fungi (yeasts and molds) in less than 5 days from a fungal culture or directly from a clinical/patient sample.
How can NGS provide unbiased detection of fungal species? The Internal Transcribed Spacer (ITS) region in the ribosomal RNA operon is conserved across fungal genomes; these conserved regions serve as primer binding sites for the amplification of the intervening sequence.

Resistance is characterized by mutations that arise under the selective pressure of antibiotics whereas antibiotic tolerance is an increased resilience to antibiotic treatment in a non-specific way.

Biofilms and tolerance are difficult to detect in traditional cultures, and without all the information, we may still be inappropriately utilizing antibiotics and contributing to the development of resistance.

A recent prospective study conducted at 14 institutions published in the Journal of Bone & Joint Surgery defines the NGS profile of 85 culture-negative PJI cases that met the ICM criteria, with NGS detecting organisms in 65.9% of culture-negative cases
