Excellent review on synthetic lethality drugs for cancer. So far only PARP and PRMT5/MAT2A inhibitors stand out as clear success stories. Most DNA damage and cell cycle checkpoints haven't panned out due to a narrow TI but there are some new promising targets (my top 3 are PARG, POLQ, WRN)
