This is HUGE: Scientists have found a way to regenerate knee cartilage and prevent arthritis.
A new study from Stanford Medicine shows that blocking an age linked protein called 15-hydroxyprostaglandin dehydrogenase (15-PGDH) can restore cartilage that normally wears away with aging.
In mice, inhibiting 15-PGDH reprogrammed existing cartilage cells (chondrocytes) into a more youthful, regenerative state. Cartilage thickness increased, joint structure improved, and inflammation dropped, without using stem cells.
The treatment also prevented osteoarthritis after ACL-like knee injuries. Injured mice given the inhibitor for about 4 weeks avoided the rapid cartilage breakdown seen in untreated animals, which typically developed arthritis within the same timeframe.
Importantly, when human knee cartilage samples from joint replacement patients were treated in the lab, researchers observed reduced cartilage degeneration gene activity and signs of renewed cartilage formation.
An oral version of the same 15-PGDH inhibitor is already in early clinical trials for age related muscle weakness, suggesting a potential path toward human therapies for joint degeneration in the future.
