Rational design of mechanically active RNAs: de novo engineering of functional exoribonuclease-resistant RNAs (xrRNAs).
We demonstrate that RNA mechanical function can be designed from first principles without relying on evolutionary sequence ancestry.
To our knowledge, this is the first example of a fully de novo engineered non-coding RNA with experimentally validated mechanical function.
Using topology- and mechanics-aware design, synthetic xrRNAs reproduce wild-type-like resistance to 5′→3′ exonucleases.
